Agrobacterium rhizogenes (inzwischen Rhizobium rhizogenes (Riker et al. 1930) Young et al. 2001[1]) ist eine Art (Spezies) pflanzenpathogener, gramnegativer Bodenbakterien. Sie gehört zur Alpha-Klasse der Proteobakterien (Alphaproteobacteria).
Rhizobium rhizogenes ist verwandt beispielsweise mit Agrobacterium tumefaciens aus derselben Familie Rhizobiaceae und hat eine ähnliche Lebensweise. Neben seiner chromosomalen DNA enthält es ein Plasmid (Ri-Plasmid). R. rhizogenes ist in der Lage, verletztes Pflanzengewebe zu infizieren und die T-DNA (Plasmid-DNA) in das pflanzliche Genom zu übertragen. Die Pflanze wird dadurch angewiesen, sogenannte Haarwurzeln auszubilden, die aus der infizierten Stelle wachsen. Je nach Bakterienstamm kann das Wurzelwachstum an fast jedem Pflanzengewebe stattfinden (zum Beispiel auch an den Blüten). Durch die Übertragung der T-DNA wird die Pflanze außerdem dazu veranlasst, Opine zu bilden, die eine Kohlenstoff- und Stickstoffquelle für das Bakterium darstellen. Die Pflanze selbst kann die Opine nicht verwerten.
Ähnlich wie bei Agrobacterium tumefaciens wird der Gen-Übertragungsmechanismus von Agrobacterium rhizogenes in der Biotechnologie zur Transformation von Pflanzen benutzt.

Die Bildung dieser Organe wird auf die Anwesenheit von dieser Bakterien zurückgeführt, deren Stamm keine IPT-Gene (verantwortlich für die Cytokinin-Synthese) in das Genom der Pflanzenzelle integriert. Eine infizierte Zelle kann daher nur in großem Umfang Auxine (wie Indolessigsäure, IAA) produzieren. Das hormonelle Gleichgewicht von IAA/CK (Auxin/Cytokinin) wird also zugunsten des Auxins verschoben, was zur Rhizogenese führt.

Cette maladie se caractérise par l'apparition d'un chevelu racinaire au point d'infection par la bactérie.
La capacité à induire la maladie est liée à la présence dans la bactérie d'un plasmide de haut poids moléculaire (ca. 300 Kb) appelé plasmide Ri (dont la structure est proche du plasmide Ti rencontré chez Agrobacterium tumefaciens), qui rend les bactéries virulentes.
Comme dans le cas de la galle du collet induite par A. tumefaciens, le hairy root résulte du transfert et de l'expression de l'information génétique portée par un fragment du plasmide pathogène, de la bactérie vers le génome nucléaire de la cellule végétale.
Cette "maladie" est en fait très utile notamment en culture végétales in vitro, afin d'induire volontairement la production de racines, à partir d'"explants" (fragments de feuilles, tiges, racines) ou de "cals" (cellules indifférenciées obtenues lors d'une étape précédente appelée callogénèse). En effet, les racines de nombreuses plantes sont capables de produire des métabolites secondaires souvent très utiles pour la production de médicaments! (Exemples : la shikonine à partir de Lithospermum erythroryzon, l'artémisinine à partir d'Artemisia annua, un antimalaria très cher à produire avec des cultures "traditionnelles"...)

Despois da integración e expresión, obsérvase, tanto in vitro coma en condicións naturais, a aparición na planta do fenotipo de raíz pilosa, o cal inclúe tipicamente un superdesenvolvemento do sistema radicular que non é completamente xeotrópico, e unha morfoloxía alterada (enrugada) das follas, se estas están presentes.[3]
Os xenes bacterianos poden ser retidos permanentemente dentro da planta, o que supón unha transferencia horizontal de xenes, que tivo importancia evolutiva.[4]
