Bacilladnaviridae ist die Bezeichnung für eine Familie von Viren mit DNA-Genom im Phylum Cressdnaviricota (en. circular REP-encoding single-stranded DNA viruses, informell CRESS-DNA-Viren). Ihre natürlichen Wirte sind hauptsächlich Kieselalgen (Diatomeen alias Bacillariophyta) wie Chaetoceros. Sie haben ein unsegmentiertes, zirkuläres Genom, das zu einem kleinen Teil Doppelstrang-DNA, ansonsten aber Einzelstrang-DNA ist.[4][5]
Die Bezeichnung Bacilladnaviridae ist zusammengesetzt aus der Bezeichnung Bacillariophyta für die Wirte, DNA, und der Endung -viridae für Virusfamilien.[6]
Die Virionen haben einen Durchmesser von etwa 34 nm – bei Chaetoceros setoensis DNA virus (CsetDNAV, Spezies Chaetoceros diatodnavirus 1) beispielsweise 33 nm[7] – und sammeln sich im Zellkern an.
Das Genom dieser Viren scheint einzigartig zu sein. Es besteht aus einem einzigen Molekül kovalent geschlossener zirkulärer einzelsträngiger DNA von 4,5–6 kb (Kilobasen) sowie (normalerweise) einem Segment (Fragment) bezeichnet linearer ssDNA von (insgesamt) ca. 0,6[7] bis 1 kb. In der Baltimore-Klassifikation weist sie dies als Mitglieder der Gruppe 2 (ssDNA-Viren) aus. Die linearen Fragmente sind aber komplementär zu einem Teil des geschlossenen Rings, wodurch in diesem teilweise doppelsträngige Regionen (dsDNA) entstehen, eine Konfiguration, die nach Baltimore nicht klassifizierbar wäre. Ausnahmen von der üblichen Zahl von einem komplementären Fragment sind beispielsweise:
● Bei Chaetoceros setoensis DNA virus (CsetDNAV) hat die zirkuläre DNA eine Länge von 5836 nt (Nukleotiden der Basen), und es gibt sogar acht kurze Segmente linearer komplementärer ssDNA mit jeweils nur 67 bis 145 nt.
● Bei dem als Mitglied vorgeschlagenen Spezies „Chaetoceros debilis DNA virus“ (CdebDNAV) gibt es offenbar keine solchen linearen Fragmente.[7]
Es gibt mindestens drei große Offene Leserahmen (englisch open reading frames, ORFs) in diesem Genom: VP1, VP2, VP3 (der Größe nach aufsteigend). D. h. es kodiert für (mindestens) drei Proteine.[6]
Wahrscheinlich replizieren Bacilladnaviridae ähnlich wie andere eukaryotische ssDNA-Viren ihr Genom per Rolling-Circle-Mechanismus, der durch die im Virusgenom kodierte Endonuklease Rep (kodiert durch VP3)[2] initiiert wird. Dieses Protein weist bei den Bacilladnaviridae jedoch unter den konservierten Sequenzmotiven für diese spezifische Besonderheiten auf und bildet in phylogenetischen Bäumen eine monophyletische Klade, die von anderen Gruppen von ssDNA-Viren getrennt ist.
Das Kapsidprotein CP (kodiert durch VP2)[2] hat eine Jelly-Roll-Faltung (englisch single jelly roll fold, [en]) und ist am engsten mit den entsprechenden Proteinen von Mitgliedern der Familie Nodaviridae [en] verwandt, die allerdings ein Einzelstrang-RNA-Genom besitzen. Offenbar ist es im Laufe der Evolution der ssDNA-Bacilladnaviridae zu einer Übernahme des Kapsid-Gens von den ssRNA-Nodaviridae gekommen.[2]
Reife Virionen werden an Ende durch Lyse der Wirtszelle freigesetzt.[6]

Bacilladnaviridae is a family of single-stranded DNA viruses that primarily infect diatoms.[1][2]
The genome of these viruses appears to be unique. It consists of a single molecule of covalently closed circular single stranded DNA of 4.5–6 kilobases as well as a segment of linear ssDNA of ~1 kilobase. The linear segment is complementary to a portion of the closed circle creating a partially double stranded region. There are at least three major open reading frames in the genome.
Similar to other eukaryotic ssDNA viruses, bacilladnaviruses are likely to replicate their genomes by the rolling-circle mechanism, initiated by the virus-encoded endonuclease (Rep). However, the latter protein of bacilladnaviruses displays unique conserved motifs and in phylogenetic trees forms a monophyletic clade separated from other groups of ssDNA viruses. The capsid protein of bacilladnaviruses has the jelly-roll fold and is most closely related to the corresponding proteins from members of the family Nodaviridae, which have ssRNA genomes.[3] [4]
The virions are about 34 nanometers (nm) in diameter and accumulate in the nucleus. Mature virions are released by lysis of the host cell.

Los virus de la familia Bacilladnaviridae tienen cápsides con geometrías icosaédricas y redondas, y simetría T = 1. No poseen envoltura vírica. El diámetro es de unos 34 nanómetros.
Los genomas son circulares y de ADN monocatenario con alrededor de 3,8 kb de longitud. Poseen un segmento lineal de ~ 1 kilobases. El segmento lineal es complementario a una porción del círculo cerrado creando una región parcial de ADN bicatenario. La cápside consta de 9 pentámeros pentagonales en forma de trompeta. Hay tres principales marcos de lectura abiertos dispuestos en direcciones opuestas que codifican las proteínas implicadas en la replicación (Rep) y la formación de la cápside (Cap).[2][1] La última proteína de los bacilladnavirus muestra motivos únicos conservados y en los árboles filogenéticos forma un clado monofilético separado de los otros virus de la clase Arfiviricetes. Según análisis filogenéticos los genes que codifica las proteínas de la cápside de los bacilladnavirus fueron adquiridos por transferencia horizontal de los virus de la familia Nodaviridae que son virus de ARN que infectan animales.[4]
La replicación viral se produce en el núcleo. La entrada en la célula huésped se logra mediante penetración en la célula huésped. La replicación sigue el modelo de círculo rodante de los virus ADN monocatenario. La transcripción con plantilla de ADN, con algún mecanismo de empalme alternativo, es el método de transcripción. Los virus salen de la célula huésped por salida nuclear y lisis. Una estructura de tallo bucle con un motivo no nucleotídico conservado se encuentra en la región intergénica 5 'de los genomas de los bacilladnavirus y se cree que inicia la replicación en círculo rodante. Las protistas sirven como hospedadores naturales. Las vías de transmisión son por reproducción y contacto.[1][2]

Le génome de ces virus est unique. Il consiste en une seule molécule d'ADN circulaire simple brin fermé de façon covalente de 4,5-6 kilobases ainsi que d'un segment d'ADN simple brin linéaire d'environ 1 kilobase. Le segment linéaire est complémentaire d'une partie de la section circulaire, ce qui crée une région partiellement double brin. Il y a au moins trois cadres de lecture ouverts majeurs dans le génome.
Comme d'autres virus à ADN simple brin, les bacilladnavirus sont susceptibles de répliquer leur génome par mécanisme de rolling circle (réplication circulaire), initiée par une endonucléase codée par le virus. Cependant, la subséquente protéine de bacilladnavirus montre la conservation de motifs uniques et, dans les arbres phylogénétiques, forme un clade (groupe monoplylétique) séparé des autres virus à ADN. La protéine de capside forme un repliement jelly roll et est plus étroitement liée aux protéines correspondantes chez les membres de la famille des Nodaviridae, des virus à ARN simple brin[5].
Les virions ont un diamètre de 34 nm et s'accumulent dans le noyau. Les virions matures sortent par lyse de la cellule infectée.
